I've been using glucosamine in combination with niacinamide as an addition to my moisturizer to combat dark spots on face and hands. Used twice daily, it does seem to keep them from darkening. Of course, you still need to be religious about using a sunscreen every time you are in the sun. .. ...»

Powered ingredient easy to blend, dissolves easily, no gritty feel, product does however transfer this chemical transdermally and should be thought about prior to mixing as it does effect hormones. The Essentials of Skin Pigmentation

 The skin is a multi-layered biological barrier that protects the internal organs and bodily structures from the continuously changing external environment, which preserves the internal stability and health of our body. Its structure mainly consists of three layers, which include the deep hypodermis layer that contains fat cells and blood vessels, the dermis that is composed of connective tissue (collagen and elastin fibers) that offer support and skin flexibility, and the epidermis that contains keratinocytes impregnated with keratin and melanocytes that produce the skin pigment melanin. The epidermis is the outermost layer, which is why it is the most important skin structure that contributes to the barrier function. The keratin deposited in the epidermic skin cells provides physical protection and resistance to the external layer of the skin, while the melanin produced in melanocytes and then transferred to keratinocytes protects against the harmful UV radiation and provides color to the skin at the same time.

Genetic and Environmental Influences of Skin Pigmentation

 Melanin is a substance that influences that color of the skin, depending on the volume and quantity of pigment granules called melanosomes, distribution throughout the surface of the skin, and the rate of melanin production and degradation. Skin color is largely influenced genetically and racially, and may range from pale, light skin in North European populations to the dark brown color in individuals originating from Africa. The differences are attributed to the amount of melanin deposited in keratinocytes and its distribution or density in the epidermis. The color of the skin can also be influenced naturally through tanning (exposure to sunlight), which stimulates the melanocytes to accelerate the rate of melanin production and transfer it to the keratinocytes located in the external layer of epidermis.

The Process of Melanin Production and Distribution

 The melanocytes are skin cells responsible for producing the melanin pigment, which protects all skin cells from damage caused by the “aggressive” ultraviolet rays that can cause skin inflammation, damage of the skin cell nuclei and alteration of skin cell structures. Melanin is therefore a protective factor against skin irritation and even tumors caused by excessive exposure to sunlight. However, acquired excessive skin pigmentation as a result of skin inflammation, skin diseases, hormonal imbalances  or physical trauma is considered unaesthetic and undesirable for most individuals. Uneven, non-uniform pigmentation is considerably more unpleasant from a cosmetic point of view than genetically determined skin pigmentation, which is why researchers have investigated potential skin care solutions to achieve a skin lightening effect.
Melanin is produced in specialized cells known as melanocytes in a cellular structure called the endoplasmic reticulum. It is then deposited in special granules called melanosomes and distributed throughout the melanocytes, including into the cell protrusions that extend far beyond the cellular body and connect each individual melanocyte with several keratinocytes. The enzyme responsible for melanin synthesis is called tyrosinase, and its stimulation (through exposure to sunlight) or inhibition (through skin lightening cosmetic products) can influence the intensity of skin pigmentation. 

Disorders that Cause Skin Hyperpigmentation and Available Cosmetic Solutions

 Besides the genetic determination and exposure to sunlight, several other factors may influence skin color and cause hyperpigmentation. Endocrine diseases (hormone imbalances), various types of nevuses, prolonged skin inflammation following trauma, tumors affecting the skin or atopic dermatitis can cause an excessive or non-uniform coloration of the skin.
Harsh  chemicals were applied in the past to achieve a rapid skin lightening effect, but the practice is associated with a high risk for skin damage and irritation. Milder and safer skin lightening agents have been explored and marketed recently, which induce a moderate and gentle inhibition of the tyrosinase enzyme and therefore limit the production of melanin without adverse effects.
